Generally speaking most digestive enzymes are destroyed by the acid the stomach unless they are enteric coated so they can pass through to the small intestine unchanged. Enzymes from raw, unprocessed foods such as papain from papaya and bromelain from pineapple and in microorganisms found supplemental as digestive aids are digested when consumed just like any other component of food. They are not active in the GI tract and they are unlikely to aid with digestion. Also, some digestive enzymes supplements contain enzymes like cellulase which helps digest cellulose but only in ruminant animals and some insect . It does not work in humans. Supplements frequently contain enzymes which the body is capable of producing in sufficient quantities without need for supplements.

Lactase and Beano are two digestive enzymes that are not destroyed by the stomach acid. Some people find they work well. Beano has not helped me.

You might find the information on Columbia University's Go Ask Alice about digestive enzyme supplements of interest and this newspaper article Enzyme hype gives some people a headache .
